Application of sodium lauryl sulfate in the field of pharmaceutical research

Sodium lauryl sulfate is a commonly used pharmaceutical excipient. It is an anionic surfactant and can be used as a solubilizer, emulsifier, skin penetrating agent, lubricant and wetting agent for tablets and capsules, etc.. The application may affect the quality, safety and effectiveness of medicines.
1.Sodium Lauryl Sulfate Basic Properties
Easily soluble in water, almost insoluble in ether and chloroform; pH value is 7.0~9.5 (10 g·L-1 aqueous solution); HLB value is about 40; under acidic conditions, it will be hydrolyzed into lauryl alcohol and sodium bisulfate, active ingredients content decreased.
2.Application in pharmaceutical preparation prescription
1)Solubilization
Amphiphilic structural compounds with different hydrophilic and hydrophobic groups in the molecule are commonly used solubilizers and co-solvents in pharmaceutical preparations. Tadalafil solid dispersion was prepared using SLS and mesoporous silica as carriers. SLS in it can significantly improve the apparent solubility and dissolution of the drug. SLS was used in the formulation of ritonavir tablets to enhance the wettability of the drug product, but the solubility of the sample in acidic media was drastically reduced. Although SLS is a conventional solubilizer, abnormal conditions also occur in different drug prescriptions, so attention should be paid to it during the drug development process.
2)Disintegration
When the tablet is made, the appropriate amount of SLS is added to improve the wettability of the tablet; the water penetration is faster, the drug dissolves faster, and the tablet disintegrates more quickly. The simultaneous use of SLS and dry starch as disintegrators is more conducive to the disintegration of tablets.
3) Antibacterial effect
SLS breaks down the cell membrane by binding to the hydrophobic part of the membrane protein and separating from the membrane. SLS in relatively high concentrations can also break non-covalent bonds in proteins and even change the conformation of proteins.
4) Lubricating effect
An appropriate amount of SLS was applied to the pharmaceutical powder mixture of celecoxib tablets, and the results showed that its lubrication efficiency was comparable to that of magnesium stearate. Still, it did not reduce the compressibility of the tablets. In addition, it was similar to the tablets containing magnesium stearate. Compared with pills, tablets containing SLS also have the effect of improving in vitro dissolution.
5)Antistatic effect
Studies have shown that SLS has antistatic capabilities and is suitable for application in pellet coating. During most of the pellet coating, the surface of pellets is in a slightly moist state, and the partially dissociated SLS has a specific conductivity, which is beneficial to eliminating static electricity. Generally, 5 to 10 g·L- is added to the coating liquid. 1 SLS can achieve the antistatic effect.
